Applications of BBa_K2263002

The Kir2.1 ion channel is very interesting for projects in the field of electrophysiology. It can be used in mammalian model systems and cells, such as HEK cells.

We have used this biobrick in our project, where we developed an electrophysiolically oscillating system in HEK cells to improve continuous drug monitoring. (Read more about that on our wiki, http://2017.igem.org/Team:KU_Leuven ) In our project, we selected three ion channels for our oscillating system based on a mathematical model. To verify that we could not just use any ion channel, we made an analog system using Kir2.1. As expected, this system did not result in oscillation. Nonetheless, the Kir2.1 ion channel might be very interesting for future projects.
We used this biobrick by cloning it into an pIRES-DSred vector, and verified its electrophysiological properties using a patch-clamp setup.
